Background

Composed of ribosomal RNA molecules and proteins, ribosomes are important tools for protein synthesis in cells. Ribosome complex profiling provides information on all ribosomes active in the cell at a specific time point and helps determine which proteins are actively translated in the cell. In life activities, mRNA translation is a key link that represents the flow of genetic information as well as defines the proteome. As an essential step in gene expression, the translation process plays important role in multiple developmental and physiological processes. To better understand translational regulation, it is important to elucidate the mechanism and regulation of translation initiation (TI).

The global translation initiation sequencing (GTI-Seq) technology has been developed to identify TIS at nucleotide resolution. The GTI-seq technology is based on the translation inhibitor lactimidomycin (LTM), which completely freezes the initiating ribosome. Different from traditional translation inhibitor cycloheximide, LTM acts preferentially on the initiation ribosome and the ribosomal footprint associated with LTM is highly enriched at annotated initiation codons. Importantly, GTI-Seq uses a simple computational approach that minimizes bias when mapping global TIS sites. To facilitate mechanistic studies of alternative translation of individual genes, a comprehensive TIS database (TISdb) based on multiple high-resolution GTI-Seq datasets has been designed.
